Description

OLA1 (Obg-like ATPase 1) encodes a member of the Obg family of P-loop GTPases/ATPases. The protein localizes to the cytoplasm and nucleus, where it functions as a translational regulator and participates in cellular responses to stress, including oxidative stress and DNA damage. OLA1 modulates the activity of the eukaryotic translation initiation factor 2 (eIF2) and interacts with heat shock proteins. It is ubiquitously expressed with highest levels in testis, heart, and skeletal muscle.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Breast cancer OLA1 overexpression promotes cell proliferation and invasion; knockdown reduces tumor growth in xenograft models PMID: 25605248
Colorectal cancer Upregulation correlates with poor prognosis; OLA1 enhances Wnt/β-catenin signaling PMID: 27794026
Lung adenocarcinoma High expression associated with metastasis and shorter survival; OLA1 regulates EMT PMID: 29367603
Hepatocellular carcinoma OLA1 silencing suppresses proliferation and induces apoptosis via p53 pathway PMID: 30351378
Neurodegenerative disorders OLA1 interacts with tau and modulates tau aggregation in cellular models PMID: 31577916

Mutation functional classification

Loss of Function (LOF)

Frameshift and nonsense mutations (e.g., p.Thr375Asnfs*2) are predicted to cause loss of ATPase activity and protein truncation.

Gain of Function (GOF)

No confirmed gain-of-function mutations reported; overexpression in tumors suggests potential oncogenic role via increased wild-type activity.

Dominant Negative (DN)

Not described for OLA1.

Pathways

• eIF2 signaling (Reactome R-HSA-156827)
• Unfolded protein response (UPR)
• p53 pathway (KEGG hsa04115)
• Wnt/β-catenin signaling (KEGG hsa04310)

Protein Summary

OLA1 is a 396-amino acid protein (molecular weight ~45 kDa) belonging to the Obg family of P-loop NTPases. It contains a conserved N-terminal GTP-binding domain and a C-terminal domain with ATPase activity. The protein shuttles between cytoplasm and nucleus, and its ATPase activity is essential for regulating translation initiation by modulating eIF2α phosphorylation. OLA1 also interacts with Hsp70/Hsp90 chaperones and is involved in maintaining proteostasis under stress conditions. Overexpression in multiple cancers suggests a role in tumor progression.
