Description

OGFR encodes the opioid growth factor receptor, a transmembrane protein that binds the opioid growth factor (OGF, Met-enkephalin). This receptor mediates the inhibitory effects of OGF on cell proliferation, particularly in epithelial and neural tissues. OGFR signaling is implicated in cell cycle regulation, tumor suppression, and wound healing. The gene is located on chromosome 20q13.33 and is expressed in a wide range of tissues.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Pancreatic Cancer OGFR downregulation reduces OGF-mediated growth inhibition, promoting tumor proliferation PMID: 19029980
Head and Neck Squamous Cell Carcinoma Reduced OGFR expression correlates with loss of OGF antiproliferative signaling PMID: 15607977
Colorectal Cancer OGFR expression is decreased in tumors, associated with poor prognosis PMID: 20043216
Corneal Wound Healing OGFR activation by OGF delays re-epithelialization; receptor blockade accelerates healing PMID: 15208134

Protein Summary

The opioid growth factor receptor (OGFR) is a 677-amino acid transmembrane protein with an extracellular domain that binds Met-enkephalin (OGF). Upon ligand binding, OGFR activates a signaling cascade that leads to upregulation of cyclin-dependent kinase inhibitors (e.g., p21, p16) and G1/S cell cycle arrest. OGFR is widely expressed in normal tissues but frequently downregulated in cancers, suggesting a tumor suppressor role. The receptor is also involved in neural development and corneal epithelial homeostasis.
