Description
OAZ1 encodes ornithine decarboxylase antizyme 1, a protein that negatively regulates polyamine biosynthesis by binding to ornithine decarboxylase (ODC1) and targeting it for ubiquitin-independent proteasomal degradation. It also inhibits polyamine uptake and is itself regulated by polyamine levels through a unique +1 ribosomal frameshift mechanism. OAZ1 plays a critical role in cell growth, differentiation, and apoptosis.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Prostate cancer | OAZ1 downregulation leads to increased ODC1 activity and polyamine accumulation, promoting cell proliferation | PMID: 10652318 |
| Colorectal cancer | Reduced OAZ1 expression correlates with elevated polyamine levels and tumor progression | PMID: 11592034 |
| Gastric cancer | OAZ1 loss contributes to ODC1 overexpression and enhanced cell growth | PMID: 15604205 |

Protein Summary
OAZ1 is a 228-amino acid protein that functions as an antizyme, inhibiting ornithine decarboxylase (ODC1) and polyamine transport. It is induced by polyamines via a ribosomal frameshift and is degraded by the proteasome. OAZ1 is widely expressed, with highest levels in testis and liver. Loss of function contributes to polyamine dysregulation in several cancers.
