Description

NPFFR2 (neuropeptide FF receptor 2) is a G protein-coupled receptor that binds neuropeptide FF (NPFF) and related peptides. It is involved in pain modulation, opioid analgesia modulation, and regulation of food intake and cardiovascular function. The receptor is expressed in the central nervous system and peripheral tissues.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Pain disorders NPFFR2 modulates nociception via Gi/o signaling; altered expression may affect pain sensitivity PMID: 12023984
Opioid tolerance NPFFR2 activation counteracts opioid-induced analgesia; chronic opioid use may upregulate NPFFR2 PMID: 15987666
Hypertension NPFFR2 expressed in cardiovascular centers; NPFF binding influences blood pressure regulation PMID: 16919457

Protein Summary

NPFFR2 is a 7-transmembrane G protein-coupled receptor of 522 amino acids. It couples primarily to Gi/o proteins, inhibiting adenylyl cyclase and modulating ion channels. The receptor is activated by neuropeptide FF (NPFF) and related RFamide peptides, playing roles in pain modulation, opioid tolerance, and cardiovascular regulation.
