Description

NMT2 encodes N-myristoyltransferase 2, an enzyme that catalyzes the covalent attachment of myristate to the N-terminal glycine residue of target proteins. This modification is essential for membrane targeting, protein-protein interactions, and signal transduction. NMT2 is ubiquitously expressed and plays roles in cellular growth, differentiation, and apoptosis. Dysregulation is linked to various cancers and infectious diseases.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Colorectal cancer Overexpression of NMT2 promotes cell proliferation and invasion via myristoylation of oncogenic proteins. PMID: 23455478
Breast cancer NMT2 upregulation correlates with poor prognosis and enhanced tumor growth. PMID: 25642768
HIV/AIDS NMT2 myristoylates HIV-1 Gag polyprotein, essential for viral assembly and infectivity. PMID: 10601273
Leishmaniasis NMT2 is a validated drug target in Leishmania species; inhibition blocks parasite growth. PMID: 15075227

Loss of Function (LOF)

Frameshift mutations (e.g., p.Leu263fs) lead to truncated, non-functional protein.

Gain of Function (GOF)

Not reported in NMT2.

• Myristoylation of HIV-1 Gag (R-HSA-162587)

Protein Summary

NMT2 is a 496-amino acid cytosolic enzyme that transfers myristate from myristoyl-CoA to the N-terminal glycine of substrate proteins. It contains a conserved catalytic domain and is essential for the function of numerous signaling and structural proteins. NMT2 is a target for anticancer and antiparasitic drug development.
