Description
NME3 encodes a member of the nucleoside diphosphate kinase (NDPK) family, specifically the NM23-H3 isoform. The protein catalyzes the transfer of gamma-phosphate from nucleoside triphosphates to nucleoside diphosphates, maintaining cellular nucleotide pools. NME3 is involved in cell proliferation, differentiation, and has been implicated in tumor metastasis suppression. It is localized to the cytoplasm and nucleus, and its expression is regulated during development and in various cancers.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Neuroblastoma | Altered NME3 expression may affect differentiation and metastatic potential; reduced expression correlates with aggressive disease. | PMID: 10652318 |
| Breast Cancer | NME3 downregulation associated with poor prognosis and increased metastatic capacity. | PMID: 12058044 |
| Hepatocellular Carcinoma | NME3 overexpression linked to tumor progression and invasion. | PMID: 17563755 |

Mutation functional classification
Loss of Function (LOF)
Mutations that reduce kinase activity or protein stability (e.g., p.Arg34Trp) may impair NME3's role in metastasis suppression.
Gain of Function (GOF)
No confirmed gain-of-function mutations reported in NME3.
Dominant Negative (DN)
Not established for NME3; dominant-negative effects have been described for other NM23 family members.

Protein Summary
NME3 is a 169-amino acid protein (18.5 kDa) belonging to the NM23 family. It forms hexamers and catalyzes the reversible transfer of the terminal phosphate between nucleoside di- and triphosphates. The protein contains a conserved NDP kinase domain and is involved in cellular energy metabolism, signal transduction, and regulation of cell motility. NME3 is ubiquitously expressed with highest levels in heart and testis. Its expression is frequently altered in cancers, where it may act as a metastasis suppressor or promoter depending on cellular context.
