Description
NME1-NME2 is a readthrough transcript that spans the NME1 and NME2 genes on chromosome 17q21.33. This transcript produces a fusion protein that combines sequences from both NME1 (nucleoside diphosphate kinase A) and NME2 (nucleoside diphosphate kinase B). The readthrough product is expressed in various tissues and is implicated in cellular energy metabolism, differentiation, and metastasis suppression. The individual NME1 and NME2 proteins are well-characterized metastasis suppressors; the readthrough isoform may have distinct or overlapping functions.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Neuroblastoma | Altered NME1/NME2 expression is associated with tumor progression and metastasis; readthrough transcript may modulate this effect. | PMID: 10647931; NCBI Gene |
| Breast carcinoma | Reduced NME1 expression correlates with high metastatic potential; readthrough transcript may influence NME1/NME2 balance. | PMID: 10647931; NCBI Gene |
| Colorectal cancer | NME1 and NME2 expression changes linked to tumor stage; readthrough transcript presence noted in some studies. | PMID: 10647931; NCBI Gene |

Protein Summary
The NME1-NME2 readthrough transcript encodes a fusion protein that combines the N-terminal region of NME1 (nucleoside diphosphate kinase A) with the C-terminal region of NME2 (nucleoside diphosphate kinase B). This protein retains nucleoside diphosphate kinase activity, catalyzing the transfer of phosphate groups between nucleoside diphosphates and triphosphates. It is involved in cellular energy homeostasis, nucleotide metabolism, and may play a role in metastasis suppression. The readthrough isoform is expressed in multiple tissues and cell lines, with highest levels in brain and heart.
