Description

NLRP10 (NLR family pyrin domain containing 10) is a member of the NLR (nucleotide-binding domain and leucine-rich repeat containing) family. Unlike most NLRs, NLRP10 lacks a leucine-rich repeat (LRR) domain and is thought to act as a negative regulator of inflammation by inhibiting ASC-dependent inflammasome activation and NF-κB signaling. It is involved in innate immune responses and epithelial barrier function.

Mutation functional classification

Loss of Function (LOF)

NLRP10 loss-of-function mutations impair negative regulation of inflammasome, leading to excessive inflammation.

Gain of Function (GOF)

No gain-of-function mutations reported.

Dominant Negative (DN)

No dominant-negative mutations reported.

Pathways

• NOD-like receptor signaling pathway (KEGG: hsa04621)
• Inflammasome pathway (Reactome: R-HSA-622312)

Protein Summary

NLRP10 is a 654-amino acid protein containing an N-terminal pyrin domain (PYD) and a central NACHT domain but lacking the C-terminal LRR domain. It functions as a negative regulator of inflammasome activation by sequestering ASC and inhibiting caspase-1 activation. Expressed mainly in epithelial tissues, NLRP10 plays a role in maintaining barrier integrity and modulating inflammatory responses.
