Description
NGB encodes neuroglobin, a member of the globin family that binds oxygen reversibly. It is predominantly expressed in the brain and retina, where it may protect neurons from hypoxic-ischemic injury. Neuroglobin is involved in oxygen transport, nitric oxide scavenging, and inhibition of apoptosis.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Alzheimer disease | Reduced NGB expression may impair neuronal oxygen homeostasis and increase oxidative stress | PMID: 20085779 |
| Ischemic stroke | NGB upregulation in response to hypoxia may limit neuronal damage | PMID: 12808427 |
| Retinal ischemia | NGB protects retinal ganglion cells from hypoxia-induced apoptosis | PMID: 15001782 |

Protein Summary
Neuroglobin is a 151-amino acid monomeric globin with a heme prosthetic group. It exhibits a typical globin fold with a distal histidine that stabilizes bound oxygen. The protein is primarily cytosolic but can translocate to the nucleus under stress. It has a higher oxygen affinity than hemoglobin and myoglobin, and its expression is induced by hypoxia via HIF-1α.
