Description
NES (nestin) encodes an intermediate filament protein expressed primarily in neural stem cells and progenitor cells during development. It is downregulated upon differentiation but re-expressed in certain cancers, particularly gliomas, where it serves as a marker of stemness and poor prognosis.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Glioma | Re-expression of nestin in glioma cells correlates with tumor grade and stem cell phenotype. | PMID: 19010870 |
| Glioblastoma | High NES expression is associated with aggressive tumor behavior and shorter survival. | PMID: 23573287 |
| Pancreatic cancer | Nestin expression in pancreatic stellate cells may promote tumor progression. | PMID: 20593220 |

Protein Summary
Nestin is a class VI intermediate filament protein of ~177 kDa. It forms heteropolymers with other intermediate filament proteins such as vimentin and internexin. Nestin is essential for the radial growth of axons and is a widely used marker for neural stem and progenitor cells. Its re-expression in various cancers, especially gliomas, makes it a potential diagnostic and prognostic biomarker.
