Description
NEDD8-MDP1 is a readthrough transcript that arises from the natural fusion of the NEDD8 (neural precursor cell expressed, developmentally down-regulated 8) and MDP1 (magnesium-dependent phosphatase 1) genes, located in close proximity on chromosome 14q11.2. This transcript is produced by the splicing of exons from both genes, resulting in a chimeric mRNA that retains the coding sequence of NEDD8 and the 3' UTR of MDP1. The readthrough event is supported by transcript evidence in Ensembl and NCBI, but no stable protein product has been experimentally validated. The transcript may play a regulatory role, potentially through the production of a NEDD8-MDP1 fusion protein or via non-coding functions, such as acting as a competitive endogenous RNA (ceRNA) or modulating the expression of its parental genes. NEDD8 is a ubiquitin-like protein involved in neddylation, a post-translational modification that regulates protein degradation and cell cycle, while MDP1 encodes a magnesium-dependent phosphatase implicated in mitochondrial function. The readthrough transcript may therefore influence these pathways, but its physiological relevance remains to be fully characterized.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Cancer (potential) | Dysregulation of NEDD8-MDP1 readthrough may affect neddylation and mitochondrial dynamics, contributing to tumorigenesis; however, direct evidence is lacking. | No direct disease association reported in ClinVar or COSMIC; inferred from parental gene functions. |
| Neurodevelopmental disorders (potential) | NEDD8 is involved in neuronal development; readthrough may alter NEDD8 expression, but no clinical evidence. | No direct evidence; speculative based on NEDD8 function. |

Protein Summary
The NEDD8-MDP1 readthrough transcript is predicted to encode a chimeric protein consisting of the N-terminal NEDD8 domain and a C-terminal portion of MDP1, but no endogenous protein has been detected. The NEDD8 domain would be expected to participate in neddylation, while the MDP1 portion may retain phosphatase activity, though the fusion may disrupt normal function. Due to lack of experimental validation, the protein's existence and function remain hypothetical.
