Description

MTA2 (Metastasis Associated 1 Family Member 2) encodes a component of the nucleosome remodeling and deacetylase (NuRD) complex. This protein functions in chromatin remodeling, histone deacetylation, and transcriptional repression. MTA2 is involved in cell cycle regulation, DNA damage response, and has been implicated in cancer metastasis and progression. It is widely expressed in various tissues and is associated with multiple malignancies.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Breast cancer Overexpression of MTA2 promotes metastasis by repressing E-cadherin and enhancing cell migration and invasion. PMID: 15694340; NCBI Gene
Gastric cancer MTA2 upregulation correlates with poor prognosis and promotes epithelial-mesenchymal transition (EMT). PMID: 23327982; NCBI Gene
Hepatocellular carcinoma MTA2 enhances tumor growth and metastasis via NuRD complex-mediated transcriptional repression. PMID: 21725308; NCBI Gene
Non-small cell lung cancer MTA2 overexpression is associated with advanced stage and lymph node metastasis. PMID: 22430205; NCBI Gene

Mutation functional classification

Loss of Function (LOF)

Frameshift and nonsense mutations that truncate the protein, disrupting NuRD complex assembly and chromatin remodeling activity.

Gain of Function (GOF)

Not well characterized; some missense mutations may enhance stability or interaction with partners, but evidence is limited.

Dominant Negative (DN)

Mutations in the SANT domain may produce a protein that competes with wild-type MTA2, impairing NuRD function.

Pathways

• NuRD complex pathway (Reactome: R-HSA-212300)
• Chromatin modifying enzymes (KEGG: hsa05200)
• Transcriptional misregulation in cancer (KEGG: hsa05202)

Protein Summary

MTA2 is a 668-amino acid protein that contains a SANT domain, a GATA-like zinc finger, and a leucine zipper motif. It is a core component of the NuRD complex, which couples ATP-dependent chromatin remodeling with histone deacetylase activity. MTA2 interacts with HDAC1/2, CHD3/4, and other NuRD subunits to repress transcription of target genes involved in cell adhesion, proliferation, and metastasis. Post-translational modifications include phosphorylation and acetylation, which modulate its stability and activity.
