Description

MST1R (Macrophage Stimulating 1 Receptor) encodes a receptor tyrosine kinase (RTK) of the MET family. It is activated by its ligand, macrophage-stimulating protein (MSP, encoded by MST1). MST1R regulates cell proliferation, migration, and survival, particularly in macrophages and epithelial cells. Aberrant expression and mutations are implicated in various cancers and inflammatory conditions.

Mutation functional classification

Loss of Function (LOF)

Rare deletions or truncating mutations reduce receptor signaling, potentially impairing macrophage function.

Gain of Function (GOF)

Missense mutations (e.g., R1024C) may enhance kinase activity, promoting oncogenic signaling.

Dominant Negative (DN)

Not well characterized; some splice variants may act as dominant negatives.

Pathways

• MSP/MST1R signaling pathway
• PI3K-Akt signaling pathway
• MAPK signaling pathway
• RAS signaling

Protein Summary

MST1R (RON) is a 180 kDa transmembrane receptor tyrosine kinase. It consists of an extracellular alpha chain and a transmembrane beta chain with intracellular kinase domain. Binding of MSP induces dimerization and autophosphorylation, activating downstream pathways such as PI3K/Akt and MAPK. Overexpression and mutations are linked to tumor progression and metastasis.
