Description

MSMP encodes beta-microseminoprotein (also known as PSP94), a small cysteine-rich secretory protein predominantly produced by the prostate gland. It is secreted into seminal plasma and is thought to play roles in sperm motility, immunomodulation, and potential tumor suppression. Reduced expression is observed in prostate cancer, suggesting a possible tumor suppressor function.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Prostate cancer Reduced MSMP expression correlates with tumor progression and poor prognosis; may act as a tumor suppressor via modulation of cell growth and apoptosis. PMID: 10652318, PMID: 15604205
Prostatic hyperplasia Altered expression levels observed in benign prostatic hyperplasia tissues compared to normal prostate. PMID: 11304735
Infertility Beta-microseminoprotein levels in seminal plasma are associated with sperm quality and fertility status. PMID: 17662314

Mutation functional classification

Loss of Function (LOF)

Loss-of-function mutations (e.g., start codon loss) are predicted to reduce or eliminate PSP94 protein, potentially contributing to prostate cancer progression.

Gain of Function (GOF)

No gain-of-function mutations have been reported for MSMP.

Dominant Negative (DN)

No dominant-negative mutations have been described for MSMP.

Pathways

• Not assigned to any specific KEGG or Reactome pathway.

Protein Summary

Beta-microseminoprotein (PSP94) is a 94-amino acid secreted protein abundant in seminal plasma. It contains 10 cysteine residues forming five disulfide bonds, characteristic of the beta-microseminoprotein family. The protein is synthesized as a precursor with a 20-amino acid signal peptide. PSP94 has been implicated in modulation of sperm motility, immunosuppression in the female reproductive tract, and potential tumor suppression in the prostate. Its expression is markedly reduced in prostate cancer tissues, and it is considered a candidate biomarker for prostate cancer diagnosis and prognosis.
