Description

MRPS28 encodes a 28S mitochondrial ribosomal protein (MRP-S28) that is a component of the small (28S) subunit of the mitochondrial ribosome. This protein is nuclear-encoded and imported into mitochondria, where it participates in mitochondrial translation. Defects in MRPS28 are associated with combined oxidative phosphorylation deficiency type 47 (COXPD47), a severe multisystem disorder characterized by lactic acidosis, developmental delay, and respiratory chain complex deficiencies.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Combined oxidative phosphorylation deficiency 47 (COXPD47) Loss-of-function mutations in MRPS28 impair mitochondrial ribosome assembly and translation, leading to reduced synthesis of mtDNA-encoded oxidative phosphorylation subunits. OMIM #619382; ClinVar; PMID: 30055966

Loss of Function (LOF)

All reported pathogenic MRPS28 mutations are loss-of-function, leading to reduced mitochondrial translation and combined OXPHOS deficiency.

Gain of Function (GOF)

No gain-of-function mutations reported.

Dominant Negative (DN)

No dominant-negative mutations reported; inheritance is autosomal recessive.

Protein Summary

MRPS28 (Mitochondrial Ribosomal Protein S28) is a 28S mitochondrial ribosomal protein encoded by the nuclear genome. It is a component of the small subunit of the mitochondrial ribosome (mitoribosome), which translates the 13 mtDNA-encoded subunits of the oxidative phosphorylation system. The protein contains a conserved ribosomal protein S28 domain and is essential for mitoribosome assembly and function. Mutations in MRPS28 cause combined oxidative phosphorylation deficiency 47 (COXPD47), an autosomal recessive disorder with severe neurological and metabolic manifestations.
