Description

MRPL17 encodes a 39S subunit protein of the mitochondrial ribosome. This protein is a component of the large ribosomal subunit (mt-LSU) and is involved in mitochondrial translation, which is essential for the synthesis of 13 subunits of the oxidative phosphorylation (OXPHOS) system. Defects in MRPL17 can impair mitochondrial protein synthesis and lead to mitochondrial dysfunction.

Disease Associations

Combined oxidative phosphorylation deficiency Loss-of-function mutations in MRPL17 impair mitochondrial translation, reducing OXPHOS complex assembly and ATP production. ClinVar, OMIM
Mitochondrial complex I deficiency Defective mitochondrial translation due to MRPL17 mutations leads to reduced complex I activity. ClinVar

Mutation functional classification

Loss of Function (LOF)

Missense and start-loss mutations reduce or abolish MRPL17 function, impairing mitochondrial translation.

Gain of Function (GOF)

No gain-of-function mutations reported.

Dominant Negative (DN)

No dominant-negative mutations reported.

• Respiratory electron transport (Reactome: R-HSA-611105)

Protein Summary

MRPL17 is a 39S mitochondrial ribosomal protein that forms part of the large ribosomal subunit. It is synthesized in the cytosol and imported into the mitochondria, where it assembles with mitochondrial rRNA and other ribosomal proteins to facilitate translation of mtDNA-encoded OXPHOS subunits. The protein contains a conserved ribosomal protein L17 domain.
