Description
MRPL12 encodes a mitochondrial ribosomal protein (L12) that is a component of the large 39S subunit of the mitochondrial ribosome. This protein is essential for mitochondrial translation, which produces 13 subunits of the oxidative phosphorylation (OXPHOS) system. MRPL12 also has a role in mitochondrial transcription regulation by interacting with mitochondrial RNA polymerase (POLRMT). Mutations in MRPL12 are associated with combined oxidative phosphorylation deficiency and mitochondrial disease.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Combined oxidative phosphorylation deficiency 36 (COXPD36) | Loss-of-function mutations impair mitochondrial translation, reducing OXPHOS complex assembly and activity. | ClinVar, OMIM #619025 |
| Mitochondrial complex I deficiency, nuclear type | Defective mitochondrial translation leads to reduced complex I subunit synthesis. | ClinVar, OMIM #252010 |
| Leigh syndrome | Severe mitochondrial dysfunction due to impaired translation can present as Leigh-like phenotype. | ClinVar, PubMed |

Protein Summary
MRPL12 is a 198-amino-acid protein (22 kDa) localized to the mitochondrial matrix. It forms part of the 39S large ribosomal subunit, interacting with rRNA and other ribosomal proteins to facilitate mRNA translation. MRPL12 also binds to POLRMT and stimulates mitochondrial transcription, linking translation and transcription. The protein contains a conserved ribosomal L12 domain and is ubiquitously expressed, with highest levels in heart and skeletal muscle.
