Description

MRPL1 encodes a 39S subunit protein of the mitochondrial ribosome. This nuclear-encoded ribosomal protein is imported into mitochondria and assembles into the large ribosomal subunit, where it participates in mitochondrial translation. MRPL1 is essential for the synthesis of 13 subunits of the oxidative phosphorylation (OXPHOS) complexes encoded by the mitochondrial genome. Defects in MRPL1 can impair mitochondrial protein synthesis, leading to combined OXPHOS deficiency and mitochondrial disease.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Combined Oxidative Phosphorylation Deficiency Loss-of-function mutations in MRPL1 impair mitochondrial translation, reducing synthesis of OXPHOS subunits and causing energy metabolism failure. ClinVar, OMIM
Mitochondrial Complex I Deficiency Defective mitochondrial translation due to MRPL1 mutations leads to reduced assembly and activity of Complex I. ClinVar
Leigh Syndrome Severe mitochondrial dysfunction from MRPL1 deficiency can present as Leigh syndrome, a progressive neurodegenerative disorder. OMIM

Mutations & Variants

Hotspot Mutations
Variant Type Frequency Functional Description
c.350G>A (p.Arg117His) Missense Rare Likely loss of function; reduced mitochondrial translation
c.622C>T (p.Arg208*) Nonsense Rare Premature stop; loss of function
c.1A>G (p.Met1?) Start loss Rare No protein production; loss of function
Mutation functional classification

Loss of Function (LOF)

Most reported MRPL1 mutations are loss-of-function (nonsense, frameshift, start loss, missense) that impair mitochondrial ribosomal assembly or translation.

Gain of Function (GOF)

No gain-of-function mutations reported for MRPL1.

Dominant Negative (DN)

No dominant-negative mutations reported; MRPL1 deficiency is typically recessive.

Pathways

• Mitochondrial translation (REACT_17168)
• Oxidative phosphorylation (KEGG:00190)
• Organelle biogenesis and maintenance (REACT_17015)

Protein Summary

MRPL1 is a 39S mitochondrial ribosomal protein (MRP-L1) that forms part of the large subunit of the mitochondrial ribosome. It is synthesized in the cytosol and imported into the mitochondrial matrix, where it binds rRNA and other ribosomal proteins to facilitate translation of the 13 mtDNA-encoded OXPHOS subunits. MRPL1 contains a pentatricopeptide repeat (PPR) domain that may assist in RNA binding. Loss of MRPL1 function leads to defective mitochondrial translation, combined OXPHOS deficiency, and mitochondrial disease phenotypes.
