Description

MORF4L1 (Mortality Factor 4 Like 1) encodes a member of the MORF4/MRG family of transcription regulatory proteins. The protein contains a chromodomain and is a component of several histone acetyltransferase (HAT) complexes, including the NuA4/TIP60 complex. It plays roles in chromatin remodeling, transcriptional activation and repression, DNA damage repair, and cell cycle regulation. MORF4L1 is ubiquitously expressed and has been implicated in cellular senescence and cancer.

Mutation functional classification

Loss of Function (LOF)

Nonsense mutations (e.g., p.Arg358*) are predicted to cause loss of function by truncating the protein.

Gain of Function (GOF)

No confirmed gain-of-function mutations reported.

Dominant Negative (DN)

No evidence for dominant-negative effects.

Pathways

• NuA4 histone acetyltransferase complex
• TIP60 complex
• DNA damage response
• Chromatin remodeling

Protein Summary

MORF4L1 (MRG15) is a 323-amino-acid protein containing an N-terminal chromodomain that mediates binding to methylated histones (e.g., H3K36me3). It is a core component of the NuA4/TIP60 HAT complex, which acetylates histones and non-histone proteins to regulate transcription, DNA repair, and cell cycle progression. The protein also interacts with other chromatin modifiers and transcription factors, influencing cellular senescence and proliferation.
