Description

MOB2 encodes a member of the MOB family of proteins that function as co-activators of NDR/LATS kinases in the Hippo signaling pathway. MOB2 specifically interacts with and activates STK38 (NDR1) and STK38L (NDR2), regulating cell cycle progression, centrosome duplication, and cell proliferation. It is involved in the control of mitotic exit and cytokinesis.

Mutation functional classification

Loss of Function (LOF)

Nonsense and frameshift mutations in MOB2 are predicted to cause loss of function, impairing NDR kinase activation and Hippo signaling.

Gain of Function (GOF)

No confirmed gain-of-function mutations reported in MOB2.

Dominant Negative (DN)

No dominant-negative mutations described for MOB2.

Pathways

• Hippo signaling pathway (KEGG: hsa04390)
• NDR kinase activation pathway
• Cell cycle regulation

Protein Summary

MOB2 is a 216-amino acid protein (25.5 kDa) that contains a conserved MOB domain. It functions as a co-activator of NDR1/NDR2 kinases, promoting their autophosphorylation and activation. MOB2 localizes to the centrosome and midbody, playing a critical role in mitotic progression and cytokinesis. Its expression is tightly regulated during the cell cycle.
