Description

MOB1B (MOB kinase activator 1B) is a protein-coding gene that encodes a component of the Hippo signaling pathway. The protein binds to and activates the serine/threonine kinases STK3 (MST2) and STK4 (MST1), which in turn phosphorylate and activate LATS1/2 kinases. This cascade regulates cell proliferation, apoptosis, and organ size. MOB1B is widely expressed and plays a role in cell cycle control and tumor suppression.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Colorectal cancer Loss of MOB1B expression leads to impaired Hippo signaling, increased YAP/TAZ activity, and enhanced cell proliferation. PMID: 25605248
Hepatocellular carcinoma Reduced MOB1B levels correlate with poor prognosis and activation of oncogenic YAP signaling. PMID: 28991257
Breast cancer MOB1B downregulation promotes cell migration and invasion via Hippo pathway dysregulation. PMID: 30108144

Protein Summary

MOB1B is a 216-amino-acid protein that functions as a co-activator of MST1/2 kinases. It contains a conserved MOB1/phocein domain and is essential for Hippo pathway activation. The protein localizes to the cytoplasm and nucleus, and its interaction with MST1/2 promotes LATS1/2 phosphorylation. MOB1B is ubiquitously expressed with highest levels in testis and brain.
