Description
MMP26 encodes a member of the matrix metalloproteinase (MMP) family, which are zinc-dependent endopeptidases involved in the degradation of extracellular matrix components. MMP26, also known as matrilysin-2 or endometase, is primarily expressed in epithelial tissues and plays a role in tissue remodeling, wound healing, and tumor invasion. It is frequently upregulated in various cancers and has been implicated in cancer progression and metastasis.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Endometrial Cancer | Overexpression of MMP26 promotes invasion and metastasis via ECM degradation. | PMID: 15604244 |
| Breast Cancer | MMP26 expression correlates with poor prognosis and increased tumor aggressiveness. | PMID: 16982746 |
| Prostate Cancer | MMP26 is upregulated and associated with advanced disease stages. | PMID: 17344455 |

Protein Summary
MMP26 is a 261-amino acid protein with a predicted molecular weight of 28.9 kDa. It contains a signal peptide, a prodomain with a cysteine switch, and a catalytic domain with the zinc-binding motif HExxHxxGxxH. Unlike other MMPs, MMP26 lacks a hemopexin-like domain. It is secreted as a zymogen and activated by proteolytic removal of the prodomain. MMP26 cleaves type IV collagen, fibronectin, and gelatin, contributing to extracellular matrix remodeling.
