Description

MMP25 (Matrix Metallopeptidase 25) encodes a member of the matrix metalloproteinase (MMP) family, specifically a membrane-type MMP (MT6-MMP). The protein is anchored to the cell membrane via a glycosylphosphatidylinositol (GPI) anchor and is involved in extracellular matrix degradation, cell migration, and inflammatory responses. It is predominantly expressed in leukocytes and plays a role in neutrophil degranulation and tumor invasion.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Colorectal Cancer Overexpression of MMP25 promotes tumor invasion and metastasis through ECM degradation and activation of pro-MMP2. PMID: 19029910
Inflammatory Bowel Disease MMP25 is upregulated in inflamed colonic mucosa and contributes to tissue remodeling and leukocyte infiltration. PMID: 16959936
Lung Cancer Elevated MMP25 expression correlates with poor prognosis and increased metastatic potential in non-small cell lung cancer. PMID: 23327982

Protein Summary

MMP25 (MT6-MMP) is a 562-amino acid membrane-anchored matrix metalloproteinase with a catalytic domain, a hinge region, and a C-terminal GPI anchor. It is synthesized as a zymogen and activated by furin cleavage. The protein degrades extracellular matrix components such as collagen IV, gelatin, and fibronectin, and activates other MMPs (e.g., pro-MMP2). It is primarily expressed in leukocytes and is implicated in inflammatory diseases and cancer metastasis.
