Description
MMP25 (Matrix Metallopeptidase 25) encodes a member of the matrix metalloproteinase (MMP) family, specifically a membrane-type MMP (MT6-MMP). The protein is anchored to the cell membrane via a glycosylphosphatidylinositol (GPI) anchor and is involved in extracellular matrix degradation, cell migration, and inflammatory responses. It is predominantly expressed in leukocytes and plays a role in neutrophil degranulation and tumor invasion.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Colorectal Cancer | Overexpression of MMP25 promotes tumor invasion and metastasis through ECM degradation and activation of pro-MMP2. | PMID: 19029910 |
| Inflammatory Bowel Disease | MMP25 is upregulated in inflamed colonic mucosa and contributes to tissue remodeling and leukocyte infiltration. | PMID: 16959936 |
| Lung Cancer | Elevated MMP25 expression correlates with poor prognosis and increased metastatic potential in non-small cell lung cancer. | PMID: 23327982 |

Protein Summary
MMP25 (MT6-MMP) is a 562-amino acid membrane-anchored matrix metalloproteinase with a catalytic domain, a hinge region, and a C-terminal GPI anchor. It is synthesized as a zymogen and activated by furin cleavage. The protein degrades extracellular matrix components such as collagen IV, gelatin, and fibronectin, and activates other MMPs (e.g., pro-MMP2). It is primarily expressed in leukocytes and is implicated in inflammatory diseases and cancer metastasis.
