Description

MMP15 encodes a member of the matrix metalloproteinase (MMP) family, specifically a membrane-type MMP (MT2-MMP). This protein is anchored to the cell membrane and functions in the degradation of extracellular matrix components, including fibronectin, laminin, and collagen types I, II, and III. It also activates pro-MMP2 and pro-MMP13, playing a key role in tissue remodeling, cell migration, and invasion. Overexpression is linked to tumor progression and metastasis in various cancers.

Loss of Function (LOF)

Nonsense mutations (e.g., c.1234C>T) are predicted to truncate the protein, reducing catalytic activity.

Gain of Function (GOF)

Not well documented; some missense variants may increase activity but evidence is limited.

Dominant Negative (DN)

No known dominant-negative mutations reported.

Protein Summary

MMP15 (MT2-MMP) is a 669-amino acid transmembrane protein with a catalytic domain, hemopexin-like domain, and a cytoplasmic tail. It is synthesized as a zymogen and activated by furin cleavage. The protein localizes to the cell surface and pericellular space, where it degrades ECM components and activates other MMPs. It is implicated in cancer cell invasion, angiogenesis, and tissue remodeling.
