Description
MMP14 encodes a membrane-type matrix metalloproteinase (MT1-MMP) that is anchored to the cell surface. It degrades extracellular matrix components (e.g., collagen I, III, fibronectin) and activates pro-MMP2. MMP14 is essential for tissue remodeling, cell migration, angiogenesis, and is implicated in cancer invasion and metastasis.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Cancer (multiple types) | Overexpression promotes ECM degradation, invasion, and metastasis via activation of MMP2 and cleavage of collagen barriers. | COSMIC, ClinVar, literature |
| Rheumatoid arthritis | Elevated MMP14 in synovial fibroblasts contributes to cartilage destruction. | OMIM, literature |
| Atherosclerosis | MMP14 expressed in macrophages and smooth muscle cells promotes plaque instability. | NCBI, literature |
| Wound healing disorders | Dysregulated MMP14 impairs proper ECM remodeling and re-epithelialization. | UniProt, literature |

Mutation functional classification
Loss of Function (LOF)
Rare missense variants near catalytic domain may impair proteolytic activity; not well characterized.
Gain of Function (GOF)
Some COSMIC-reported missense variants (e.g., p.Gly349Arg) are suspected to enhance activity in cancer cells.
Dominant Negative (DN)
Not reported for MMP14.

Protein Summary
MMP14 (MT1-MMP) is a 582-amino acid transmembrane metalloproteinase with a catalytic domain, hemopexin-like domain, and a cytoplasmic tail. It localizes to the cell surface and pericellular space, where it degrades collagen I, III, fibronectin, and activates pro-MMP2. It is a key driver of cell invasion, angiogenesis, and tumor metastasis. Its expression is upregulated in many cancers and inflammatory diseases.
