Description

MATK (megakaryocyte-associated tyrosine kinase) encodes a non-receptor tyrosine kinase belonging to the CSK family. The protein negatively regulates Src family kinases by phosphorylating their C-terminal tyrosine residue, thereby modulating signal transduction pathways involved in hematopoiesis, immune cell activation, and cell proliferation. MATK is predominantly expressed in hematopoietic tissues, particularly megakaryocytes and T cells.

Mutation functional classification

Loss of Function (LOF)

Frameshift and nonsense mutations predicted to truncate the kinase domain, impairing negative regulation of Src kinases.

Gain of Function (GOF)

Not well characterized; rare missense variants may alter substrate specificity.

Dominant Negative (DN)

No evidence currently available.

Pathways

• T-cell receptor signaling pathway (KEGG: hsa04660)
• Fc epsilon RI signaling pathway (KEGG: hsa04664)
• Natural killer cell mediated cytotoxicity (KEGG: hsa04650)
• Regulation of Src family kinases (Reactome: R-HSA-912526)

Protein Summary

The MATK protein (42679 Da) is a cytoplasmic tyrosine kinase containing SH2, SH3, and catalytic domains. It phosphorylates the C-terminal regulatory tyrosine of Src family kinases (e.g., SRC, LCK, FYN), leading to their inactivation. This negative regulation is critical for controlling immune cell activation and hematopoietic differentiation. MATK is also known as CSK-type kinase (CTK) or hematopoietic consensus kinase (CHK).
