Description
The MAS1 gene encodes a G protein-coupled receptor (GPCR) that functions as a receptor for angiotensin-(1-7), a vasoactive peptide of the renin-angiotensin system. It mediates vasodilation, anti-proliferative, and anti-inflammatory effects, counterbalancing the actions of the angiotensin II type 1 receptor. MAS1 is involved in cardiovascular regulation, neuroprotection, and metabolic homeostasis. It was originally identified as a proto-oncogene from a human epidermoid carcinoma cell line.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Cardiovascular disease | MAS1 signaling via angiotensin-(1-7) promotes vasodilation and reduces blood pressure; dysregulation contributes to hypertension and heart failure | ClinVar, OMIM |
| Neurological disorders | MAS1 activation exerts neuroprotective effects; altered expression linked to Alzheimer's disease and stroke | NCBI Gene, OMIM |
| Metabolic syndrome | MAS1 modulates insulin sensitivity and glucose metabolism; deficiency associated with insulin resistance | UniProt, OMIM |

Protein Summary
The MAS1 protein is a 325-amino acid G protein-coupled receptor with seven transmembrane domains. It is activated by angiotensin-(1-7) and couples primarily to Gαi/o proteins, leading to inhibition of adenylyl cyclase and activation of downstream kinases such as PI3K/Akt and MAPK. MAS1 is expressed in brain, heart, kidney, and testis, where it mediates vasodilation, anti-fibrotic, and anti-inflammatory effects. It plays a protective role in cardiovascular and neurological diseases.
