Description

MAPRE1 (Microtubule Associated Protein RP/EB Family Member 1), commonly known as EB1, encodes a member of the RP/EB family of microtubule-associated proteins. This protein localizes to the plus ends of microtubules and regulates microtubule dynamics, cell polarity, and chromosome stability. It interacts with the adenomatous polyposis coli (APC) tumor suppressor and is involved in cell migration and division.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Colorectal Cancer MAPRE1 overexpression stabilizes microtubule plus ends, promoting cell migration and invasion; interacts with APC in Wnt signaling. PMID: 12058064, COSMIC
Breast Cancer Elevated MAPRE1 expression correlates with poor prognosis; enhances microtubule dynamics and metastasis. PMID: 22926525, COSMIC
Ciliopathies MAPRE1 localizes to centrosomes and cilia; mutations disrupt ciliary microtubule organization. PMID: 20089971, ClinVar

Mutations & Variants

Hotspot Mutations
Variant Type Frequency Functional Description
c.1A>G (p.Met1?) Missense <0.1% Loss of start codon; likely loss of function
c.487C>T (p.Arg163Trp) Missense <0.1% Alters EB1-APC interaction; reported in ClinVar

Mutation functional classification

Loss of Function (LOF)

Rare truncating or start-loss mutations (e.g., p.Met1?) reduce protein expression or disrupt microtubule binding.

Gain of Function (GOF)

Overexpression in cancers (e.g., colorectal, breast) enhances microtubule stability and cell motility; no activating point mutations reported.

Dominant Negative (DN)

Mutations in the EB1-APC interaction domain (e.g., p.Arg163Trp) may disrupt APC-mediated regulation, acting in a dominant-negative manner.

Pathways

Regulation of microtubule dynamics (Reactome: R-HSA-983231)
APC/C-mediated degradation of cell cycle proteins (Reactome: R-HSA-174178)
Wnt signaling pathway (KEGG: hsa04310)

Protein Summary

MAPRE1 (EB1) is a 268-amino-acid protein that binds to the plus ends of growing microtubules via its N-terminal calponin homology domain. It recruits various interacting partners (e.g., APC, CLIP-170) to regulate microtubule dynamics, spindle positioning, and cell polarity. The C-terminal EB1 domain mediates dimerization and interactions with other plus-end tracking proteins. Post-translational modifications include phosphorylation and acetylation, which modulate its localization and function.
