Description

MAPKAPK5 (MAPK activated protein kinase 5), also known as PRAK (p38-regulated/activated protein kinase), is a serine/threonine kinase that acts downstream of p38 MAPK signaling. It plays roles in stress-induced responses, cell cycle regulation, senescence, and tumor suppression. MAPKAPK5 phosphorylates substrates such as HSP27 and regulates gene expression. It is widely expressed and implicated in various cancers and inflammatory conditions.

Mutation functional classification

Loss of Function (LOF)

Frameshift or truncating mutations that abolish kinase activity are likely loss-of-function.

Gain of Function (GOF)

No clear gain-of-function mutations reported; overexpression may mimic activation.

Dominant Negative (DN)

Kinase-dead mutants (e.g., ATP-binding site mutations) can act as dominant-negative in experimental settings.

Protein Summary

MAPKAPK5 is a 473-amino acid protein with an N-terminal kinase domain and a C-terminal regulatory region. It is activated by p38 MAPK-mediated phosphorylation at Thr182. It phosphorylates substrates like HSP27 and regulates actin dynamics. It is involved in cell proliferation, differentiation, and stress responses. Subcellular localization is predominantly nuclear and cytoplasmic.
