Description

MAPKAPK3 (Mitogen-Activated Protein Kinase-Activated Protein Kinase 3) encodes a serine/threonine protein kinase that is activated by p38 MAPK and to a lesser extent by ERK. It phosphorylates downstream targets involved in stress response, inflammation, cell cycle regulation, and cytoskeletal remodeling. The gene is located on chromosome 3p21.31 and is widely expressed.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Cancer (various) MAPKAPK3 is implicated in tumorigenesis via p38 MAPK signaling; altered expression affects cell proliferation and apoptosis. COSMIC, NCBI
Inflammatory disorders MAPKAPK3 regulates cytokine production (e.g., TNF, IL-6) through phosphorylation of transcription factors. UniProt, NCBI
Cardiovascular disease MAPKAPK3 modulates cardiac hypertrophy and stress responses via p38 pathway. NCBI, OMIM

Loss of Function (LOF)

Frameshift mutations (e.g., c.788_789insA) predicted to truncate the kinase domain, reducing catalytic activity.

Gain of Function (GOF)

No confirmed gain-of-function mutations reported in major databases.

Protein Summary

MAPKAPK3 is a 382-amino acid serine/threonine kinase (UniProt Q16644) with a molecular weight of ~43 kDa. It contains an N-terminal kinase domain and a C-terminal regulatory region. Activation occurs via phosphorylation by p38 MAPK at Thr222 and Ser272. The kinase phosphorylates substrates such as HSP27, CREB, and SRF, influencing stress responses, cytoskeletal dynamics, and gene expression. It is ubiquitously expressed with highest levels in testis and brain.
