Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Cancer (various) MAPK7 overexpression or constitutive activation promotes tumor growth and metastasis via transcriptional regulation of c-Jun, MEF2, and other targets. COSMIC; PMID: 25605248
Cardiovascular defects MAPK7 knockout mice show embryonic lethality due to impaired cardiac development and vascular integrity. OMIM #602521; PMID: 10572027

Pathways

MAPK7 (mitogen-activated protein kinase 7)
also known as ERK5 or BMK1
encodes a member of the MAP kinase family. This kinase is activated by upstream kinases (MEK5) in response to growth factors and stress stimuli
and phosphorylates downstream targets involved in cell proliferation
differentiation
survival
and angiogenesis. MAPK7 is essential for cardiovascular development and has been implicated in various cancers and neurodegenerative diseases.
MAPK signaling pathway (KEGG: hsa04010)
ErbB signaling pathway (KEGG: hsa04012)
Neurotrophin signaling pathway (KEGG: hsa04722)
MEK5-ERK5 signaling
MAPK7 (ERK5) is a 816-amino acid serine/threonine kinase with an N-terminal kinase domain and a unique C-terminal tail containing nuclear localization and transactivation domains. It is activated by MEK5 via dual phosphorylation on Thr218 and Tyr220. ERK5 regulates gene expression through phosphorylation of transcription factors such as MEF2
c-Jun
and Sap1a
and is critical for cell cycle progression
survival
and angiogenesis.
