Description

MAPK3 (Mitogen-Activated Protein Kinase 3) encodes a member of the MAP kinase family, also known as extracellular signal-regulated kinase 1 (ERK1). This protein integrates signals from growth factors, cytokines, and stress stimuli, phosphorylating downstream targets to regulate cell cycle progression, differentiation, transcription, and apoptosis. MAPK3 is ubiquitously expressed and is a central node in the RAS-RAF-MEK-ERK cascade.

Protein Summary

MAPK3 (ERK1) is a 44 kDa serine/threonine kinase that, together with ERK2, mediates signal transduction from cell surface receptors to the nucleus. It is activated by MEK1/2 via dual phosphorylation on Thr202 and Tyr204. ERK1 phosphorylates hundreds of substrates including transcription factors (ELK1, c-Fos), kinases (RSK), and cytoskeletal proteins. Its activity is tightly regulated; dysregulation is a hallmark of many cancers and developmental disorders.
