MAPK14 (p38α) is a 360-amino-acid serine/threonine kinase with a conserved dual phosphorylation site (TGY motif) in the activation loop. It is ubiquitously expressed and localizes to both cytoplasm and nucleus. Upon activation by upstream kinases (MKK3/6)
p38α phosphorylates numerous substrates including transcription factors (ATF2
MEF2C)
kinases (MAPKAPK2)
and apoptotic regulators. Its structure includes an N-terminal lobe (β-sheets) and a C-terminal lobe (α-helices) forming the ATP-binding cleft. Dysregulation of MAPK14 is implicated in chronic inflammation
neurodegenerative diseases
and multiple cancers.
