Description
MAPK11 (Mitogen-Activated Protein Kinase 11) encodes a member of the p38 mitogen-activated protein kinase (MAPK) family, specifically p38 beta. This serine/threonine kinase is activated by various environmental stresses and pro-inflammatory cytokines through a phosphorylation cascade involving MAP2Ks (e.g., MKK3, MKK6). p38 beta shares high sequence similarity with p38 alpha (MAPK14) but exhibits distinct substrate preferences and regulatory roles. MAPK11 is involved in cell differentiation, apoptosis, autophagy, and inflammatory responses. It is widely expressed, with highest levels in heart, brain, and skeletal muscle.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Cancer (various) | Dysregulated p38 signaling can promote or suppress tumorigenesis depending on context; MAPK11 mutations may alter kinase activity affecting cell proliferation and survival. | COSMIC; literature review |
| Inflammatory disorders | MAPK11 mediates cytokine production (e.g., TNF-alpha, IL-1) and immune cell activation; aberrant activation contributes to chronic inflammation. | UniProt; PubMed |
| Neurodegenerative diseases | p38 beta is implicated in neuronal stress responses; altered MAPK11 activity may influence tau phosphorylation and neuroinflammation. | OMIM; literature |

Mutation functional classification
Loss of Function (LOF)
Missense mutations in the kinase domain (e.g., Thr38Ile) may reduce catalytic activity, impairing downstream signaling.
Gain of Function (GOF)
No confirmed gain-of-function mutations reported in MAPK11.
Dominant Negative (DN)
Kinase-dead mutants can act as dominant negatives by sequestering upstream activators or competing with wild-type p38.

Protein Summary
MAPK11 (p38 beta) is a 364-amino-acid serine/threonine kinase with a conserved dual phosphorylation motif (TGY). It is activated by MKK3/MKK6 in response to stress and cytokines. p38 beta phosphorylates downstream targets including transcription factors (ATF2, MEF2C), kinases (MAPKAPK2/3), and other substrates involved in mRNA stability, cytoskeletal dynamics, and cell cycle control. Unlike p38 alpha, p38 beta has lower affinity for SB203580 inhibitor, suggesting distinct pharmacological profiles. The protein is predominantly cytoplasmic but can translocate to the nucleus upon activation.
