Description

MAPK11 (Mitogen-Activated Protein Kinase 11) encodes a member of the p38 mitogen-activated protein kinase (MAPK) family, specifically p38 beta. This serine/threonine kinase is activated by various environmental stresses and pro-inflammatory cytokines through a phosphorylation cascade involving MAP2Ks (e.g., MKK3, MKK6). p38 beta shares high sequence similarity with p38 alpha (MAPK14) but exhibits distinct substrate preferences and regulatory roles. MAPK11 is involved in cell differentiation, apoptosis, autophagy, and inflammatory responses. It is widely expressed, with highest levels in heart, brain, and skeletal muscle.

Protein Summary

MAPK11 (p38 beta) is a 364-amino-acid serine/threonine kinase with a conserved dual phosphorylation motif (TGY). It is activated by MKK3/MKK6 in response to stress and cytokines. p38 beta phosphorylates downstream targets including transcription factors (ATF2, MEF2C), kinases (MAPKAPK2/3), and other substrates involved in mRNA stability, cytoskeletal dynamics, and cell cycle control. Unlike p38 alpha, p38 beta has lower affinity for SB203580 inhibitor, suggesting distinct pharmacological profiles. The protein is predominantly cytoplasmic but can translocate to the nucleus upon activation.
