Description
MAP3K12 (Mitogen-Activated Protein Kinase Kinase Kinase 12) encodes a member of the serine/threonine protein kinase family. This kinase specifically activates the JNK (c-Jun N-terminal kinase) signaling pathway by phosphorylating and activating MAP2K4/MKK4 and MAP2K7/MKK7. MAP3K12 plays a critical role in neuronal development, stress response, and apoptosis. It is also implicated in neurodegenerative diseases and certain cancers.

Mutation functional classification
Loss of Function (LOF)
Nonsense and frameshift mutations that truncate the kinase domain, impairing JNK activation and neuronal survival.
Gain of Function (GOF)
Not well documented; some missense variants may increase kinase activity but evidence is limited.
Dominant Negative (DN)
Truncated forms lacking the C-terminal regulatory domain may interfere with wild-type MAP3K12 signaling.

Protein Summary
MAP3K12 is a 954-amino acid serine/threonine kinase with an N-terminal kinase domain and a C-terminal leucine zipper domain. It acts as a key activator of the JNK pathway, regulating stress responses, neuronal differentiation, and apoptosis. The protein is predominantly expressed in the nervous system and testis.
