Description

MAP2K5 (MEK5) is a dual-specificity protein kinase that specifically phosphorylates and activates MAPK7 (ERK5). It functions in the MAPK signaling cascade, regulating cell proliferation, differentiation, and survival. MAP2K5 is activated by upstream kinases such as MAP3K2/3 and is involved in neurodevelopment, cardiac function, and cancer progression.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Cancer (various) MAP2K5 overexpression or activating mutations drive ERK5 signaling, promoting proliferation and metastasis. COSMIC, ClinVar
Neurodevelopmental disorders Missense variants in MAP2K5 have been associated with intellectual disability and developmental delay. ClinVar, OMIM
Cardiac hypertrophy MAP2K5-ERK5 pathway is implicated in cardiomyocyte growth and stress responses. NCBI Gene, PubMed

Loss of Function (LOF)

Rare missense variants (e.g., p.Arg339Trp) may reduce kinase activity, linked to neurodevelopmental phenotypes.

Gain of Function (GOF)

Amplification or activating mutations (e.g., p.Glu416Lys) enhance ERK5 signaling, promoting oncogenesis.

Dominant Negative (DN)

Not well characterized; some engineered mutants can inhibit ERK5 activation.

Gene Ontology (GO)

• MAP kinase kinase activity • protein serine/threonine kinase activity
• ATP binding • signal transduction
• MAPK cascade • regulation of cell proliferation

Pathways

• ERK5 signaling (Reactome: R-HSA-198753)

Protein Summary

MAP2K5 (MEK5) is a 448-amino acid dual-specificity kinase that phosphorylates ERK5 on Thr218/Tyr220. It contains an N-terminal PB1 domain for interaction with upstream MAP3Ks and a C-terminal kinase domain. MEK5 is critical for ERK5-mediated transcription and cellular responses to growth factors and stress.
