Protein Summary

MAP1LC3B encodes the LC3B protein, a member of the ATG8 family. The protein is synthesized as pro-LC3B, which is cleaved by ATG4 to form cytosolic LC3-I. Upon autophagy induction, LC3-I is conjugated to phosphatidylethanolamine by ATG7, ATG3, and the ATG12-ATG5-ATG16L1 complex to generate lipidated LC3-II, which associates with autophagosomal membranes. LC3-II is a widely used marker for autophagic activity. The protein also interacts with microtubules and actin, linking autophagy to cytoskeletal dynamics. Post-translational modifications include lipidation, acetylation, and ubiquitination, which regulate its function.
