Description

MAGEA11 is a member of the melanoma-associated antigen (MAGE) gene family, which encodes cancer-testis antigens (CTAs) that are normally expressed in germ cells but aberrantly expressed in various cancers. MAGEA11 functions as a transcriptional co-regulator, interacting with androgen receptor (AR) and other nuclear receptors to modulate gene expression. It is implicated in tumorigenesis, particularly in prostate and breast cancers, and may serve as a target for immunotherapy.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Prostate cancer MAGEA11 enhances androgen receptor transcriptional activity, promoting tumor growth PMID: 19029910
Breast cancer Overexpression of MAGEA11 correlates with poor prognosis and hormone receptor status PMID: 23382299
Melanoma Aberrant expression as a cancer-testis antigen; potential immunotherapeutic target PMID: 10557338

Protein Summary

MAGEA11 is a 429-amino acid protein belonging to the MAGE family. It contains a conserved MAGE homology domain (MHD) and functions as a co-regulator of androgen receptor and other nuclear receptors. The protein is localized to the nucleus and modulates transcription by recruiting coactivators or corepressors. Its expression is typically restricted to testis and placenta but is reactivated in various cancers, contributing to tumor progression.
