Description

MAFG (MAF bZIP transcription factor G) is a member of the MAF family of basic leucine zipper (bZIP) transcription factors. It forms homodimers or heterodimers with other bZIP proteins, such as NFE2, to regulate gene expression. MAFG is involved in cellular stress responses, differentiation, and hematopoiesis. It plays a role in the antioxidant response by binding to antioxidant response elements (AREs) in the promoters of target genes.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Cancer (various types) MAFG overexpression or mutation may alter transcriptional programs affecting cell proliferation and survival. COSMIC; PMID: 25691885
Hereditary elliptocytosis Mutations in MAFG have been associated with red blood cell membrane disorders. OMIM #602020; PMID: 10447258
Oxidative stress-related disorders MAFG regulates antioxidant genes via ARE; dysregulation contributes to disease. NCBI Gene; PMID: 12529312

Mutation functional classification

Loss of Function (LOF)

Frameshift and nonsense mutations are predicted to result in loss of function.

Gain of Function (GOF)

Not well characterized; some missense variants may enhance transcriptional activity.

Dominant Negative (DN)

Heterodimerization with other bZIP factors could be disrupted by certain mutations.

Pathways

• Antioxidant response (ARE) pathway
• NFE2/MAF signaling in erythropoiesis
• Cellular stress response

Protein Summary

MAFG is a 162-amino acid protein containing a bZIP domain that mediates dimerization and DNA binding. It is predominantly nuclear and functions as a transcriptional activator or repressor depending on its dimerization partner. MAFG is critical for the expression of phase II detoxifying enzymes and antioxidant proteins, and it plays a role in erythroid differentiation.
