Description

LY6E encodes a glycosylphosphatidylinositol (GPI)-anchored cell surface protein belonging to the Ly6/uPAR family. It is involved in immune cell regulation, T cell activation, and modulation of viral entry. LY6E is highly expressed in hematopoietic tissues and is upregulated in response to interferons. It has been identified as a host factor that restricts infection by enveloped viruses, including SARS-CoV-2, influenza A, and HIV-1.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
COVID-19 LY6E restricts SARS-CoV-2 entry by inhibiting membrane fusion PMID: 32818467
Influenza A LY6E blocks influenza A virus entry at the fusion step PMID: 28325809
HIV-1 LY6E inhibits HIV-1 infection by interfering with viral fusion PMID: 28325809
Breast cancer Overexpression of LY6E correlates with poor prognosis and metastasis PMID: 25609812
Acute myeloid leukemia LY6E is upregulated in AML and associated with leukemic stem cell markers PMID: 21931115

Protein Summary

LY6E is a small, GPI-anchored cell surface protein (131 amino acids) with a single LU domain. It is expressed on hematopoietic cells and upregulated by interferons. The protein localizes to the plasma membrane and endosomes, where it inhibits viral membrane fusion. In cancer, LY6E promotes cell migration and invasion, and its overexpression is linked to poor prognosis in breast cancer and leukemia.
