Description

LTA (lymphotoxin alpha) is a member of the tumor necrosis factor (TNF) superfamily. It encodes a cytokine produced by lymphocytes that mediates inflammatory and immune responses. LTA forms heterotrimers with lymphotoxin beta (LTB) to activate NF-kappaB pathways, playing roles in lymphoid organ development, apoptosis, and host defense. Genetic variants in LTA are associated with susceptibility to myocardial infarction, asthma, and autoimmune diseases.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Myocardial Infarction LTA polymorphisms (e.g., rs909253) alter cytokine expression, promoting vascular inflammation and atherosclerosis NCBI Gene, OMIM
Asthma LTA variants influence airway inflammation and immune hyperresponsiveness NCBI Gene, OMIM
Rheumatoid Arthritis LTA contributes to synovial inflammation via TNF pathway activation NCBI Gene, OMIM
Systemic Lupus Erythematosus LTA dysregulation linked to autoantibody production and immune complex deposition NCBI Gene, OMIM

Mutation functional classification

Loss of Function (LOF)

Rare start codon mutations (e.g., c.1A>G) predicted to abolish protein expression.

Gain of Function (GOF)

Not well documented; some SNPs (e.g., rs909253) may increase LTA transcription.

Dominant Negative (DN)

Not reported for LTA.

Pathways

TNF signaling pathway (KEGG hsa04668)
NF-kappa B signaling pathway (KEGG hsa04064)
Cytokine-cytokine receptor interaction (KEGG hsa04060)

Protein Summary

Lymphotoxin alpha (LTA) is a 25 kDa secreted cytokine of the TNF superfamily. It is primarily produced by activated T and B lymphocytes. LTA can form soluble homotrimers or membrane-bound heterotrimers with LTB. The LTA-LTB complex binds to LTBR (lymphotoxin beta receptor) to activate NF-kappaB and MAPK pathways, regulating lymphoid organogenesis, inflammation, and cell survival. LTA is implicated in atherosclerosis, asthma, and autoimmune diseases.
