Description
LSM10 encodes a protein that is a component of the U7 small nuclear ribonucleoprotein (snRNP) complex. This complex is essential for the 3' end processing of replication-dependent histone pre-mRNAs, which is a critical step in cell cycle-regulated histone mRNA maturation. LSM10 is a homolog of the Sm proteins and is specifically associated with the U7 snRNA, contributing to the unique function of the U7 snRNP in histone mRNA processing.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Disease | Mechanism | Evidence |
| Cancer (various types) | Altered expression of LSM10 may disrupt histone mRNA processing, affecting cell cycle progression and genomic stability. | COSMIC database lists LSM10 mutations in multiple cancer types; expression changes observed in some studies. |
| Developmental disorders (potential) | Given its role in histone mRNA processing, mutations could impair cell proliferation during development. | No direct OMIM disease association; limited evidence from model organisms. |

Hotspot Mutations
| Variant | Type | Frequency | Functional Description |
|---|---|---|---|
| Variant | Type | Frequency | Effect |
| c.1A>G (p.Met1Val) | Missense | Rare (<0.01%) | Potential loss of start codon, leading to truncated or absent protein. |
| c.214C>T (p.Arg72Trp) | Missense | Rare (<0.01%) | May affect protein stability or interaction with U7 snRNA. |
| c.315_316del (p.Glu106fs) | Frameshift | Rare (<0.01%) | Predicted to cause premature termination, likely loss of function. |
Mutation functional classification
Loss of Function (LOF)
Frameshift and nonsense mutations that truncate the protein are likely to result in loss of function, impairing U7 snRNP assembly and histone mRNA processing.
Gain of Function (GOF)
No evidence for gain-of-function mutations; LSM10 is not known to have oncogenic activity.
Dominant Negative (DN)
Missense mutations in the Sm domain could potentially exert dominant-negative effects by interfering with U7 snRNP assembly, but this is speculative.

Protein Summary
LSM10 is a 100-amino acid protein that belongs to the Sm-like protein family. It forms a heterodimer with LSM11, which is a core component of the U7 snRNP. The U7 snRNP recognizes the histone downstream element (HDE) in replication-dependent histone pre-mRNAs and recruits the cleavage and polyadenylation specificity factor (CPSF) complex to mediate 3' end cleavage. LSM10 is essential for the stability and function of the U7 snRNP. Its expression is cell cycle-regulated, peaking during S phase when histone mRNA synthesis is highest.
