Description

LRP10 (LDL Receptor Related Protein 10) encodes a member of the low-density lipoprotein (LDL) receptor family. The protein functions as an endocytic receptor involved in the cellular uptake of ligands, including apolipoprotein E (ApoE) and amyloid precursor protein (APP). LRP10 is widely expressed, with highest levels in the brain, and plays a critical role in lipid metabolism, neuronal signaling, and the clearance of amyloid-beta peptides. Genetic variants in LRP10 have been associated with an increased risk of Alzheimer's disease and Parkinson's disease.

Mutation functional classification

Loss of Function (LOF)

Missense variants (e.g., p.Arg48Cys, p.Arg510Cys) reduce receptor internalization and ligand clearance, leading to amyloid-beta accumulation.

Gain of Function (GOF)

No gain-of-function mutations have been reported for LRP10.

Dominant Negative (DN)

Heterozygous missense mutations may exert dominant-negative effects by forming non-functional dimers with wild-type LRP10.

Pathways

• Alzheimer disease - amyloid-beta clearance pathway (Reactome: R-HSA-977225)
• LDL receptor-mediated endocytosis (Reactome: R-HSA-8866423)
• Lipoprotein metabolism (KEGG: hsa04979)

Protein Summary

LRP10 is a 777-amino acid transmembrane protein belonging to the LDL receptor family. It contains ligand-binding type A repeats, an EGF-like domain, a transmembrane domain, and a cytoplasmic NPxY motif essential for endocytosis. The protein is primarily localized to the plasma membrane and endosomes, where it mediates the internalization of ligands such as ApoE and APP. LRP10 is highly expressed in neurons and glial cells, and its dysfunction is linked to impaired amyloid-beta clearance and neurodegeneration.
