Description

LMAN2 (Lectin, Mannose Binding 2), also known as VIP36, encodes a type I transmembrane lectin that functions as a cargo receptor in the early secretory pathway. It cycles between the endoplasmic reticulum (ER) and the Golgi apparatus, binding high-mannose-type glycans to facilitate the transport and quality control of glycoproteins. LMAN2 is ubiquitously expressed and plays a role in maintaining ER homeostasis and protein secretion.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Congenital disorders of glycosylation (CDG) LMAN2 mutations may impair glycoprotein trafficking, leading to defective glycosylation patterns. UniProt; OMIM
Cancer (various) Altered LMAN2 expression has been observed in several cancers, potentially affecting tumor cell glycoprotein secretion and immune evasion. COSMIC; NCBI Gene

• Quality control of glycoproteins (UniProt)

Protein Summary

LMAN2 (VIP36) is a 36 kDa type I transmembrane lectin localized to the ER-Golgi intermediate compartment. It binds high-mannose oligosaccharides on glycoproteins, acting as a cargo receptor for their transport from the ER to the Golgi. The protein contains a luminal carbohydrate recognition domain, a transmembrane region, and a short cytoplasmic tail. It cycles between compartments via COPI and COPII vesicles, contributing to glycoprotein quality control and secretion.
