Description

LIN28B encodes a RNA-binding protein that inhibits the maturation of let-7 family microRNAs, thereby promoting stem cell self-renewal and proliferation. It is involved in developmental timing, glucose metabolism, and is frequently overexpressed in various cancers, contributing to poor prognosis and metastasis.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Hepatocellular carcinoma LIN28B overexpression suppresses let-7, leading to de-repression of oncogenes (e.g., HMGA2, MYC) and enhanced proliferation PMID: 21499247
Colorectal cancer LIN28B amplification and overexpression correlate with advanced stage and poor survival via let-7 downregulation PMID: 21746835
Ovarian cancer LIN28B upregulation promotes tumorigenesis and chemoresistance through let-7 inhibition PMID: 22949634
Wilms tumor LIN28B overexpression is associated with aggressive histology and relapse PMID: 25261932
Type 2 diabetes LIN28B variants influence insulin sensitivity and glucose homeostasis via let-7 regulation PMID: 25985138

Mutations & Variants

Hotspot Mutations
Variant Type Frequency Functional Description
c.1A>G (p.Met1?) Missense <0.01% Loss of start codon; likely loss of function
c.200C>T (p.Pro67Leu) Missense <0.01% Unknown significance
c.500G>A (p.Arg167Gln) Missense <0.01% Unknown significance

Mutation functional classification

Loss of Function (LOF)

Rare truncating or start-loss variants (e.g., p.Met1?) are predicted to abolish protein function.

Gain of Function (GOF)

Gene amplification and overexpression in multiple cancers lead to increased suppression of let-7 and oncogenic activation.

Dominant Negative (DN)

No dominant-negative mutations have been reported for LIN28B.

Pathways

• let-7 inhibition of oncogenes (HMGA2
• MYC
• RAS)
• LIN28/let-7 pathway in stem cell differentiation
• PI3K/AKT signaling (via let-7 targets)
• p53 pathway (via let-7 regulation of CDKN1A)

Protein Summary

LIN28B is a 250-amino acid RNA-binding protein containing a cold-shock domain and two CCHC zinc finger motifs. It binds to the terminal loop of let-7 precursor microRNAs, blocking their processing by DICER and DROSHA. This leads to reduced mature let-7 levels, allowing expression of oncogenic targets such as HMGA2, MYC, and RAS. LIN28B also modulates mRNA translation and stability independently of let-7. Its expression is high in embryonic stem cells and certain cancers, where it promotes self-renewal, proliferation, and metabolic reprogramming.
