Description
The LIF gene encodes leukemia inhibitory factor, a pleiotropic cytokine of the interleukin-6 family. It regulates cell growth, differentiation, and survival in various tissues, including embryonic stem cells, hematopoietic cells, and neurons. LIF is involved in implantation, inflammation, and cancer progression.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Cancer (various) | LIF promotes tumor growth and metastasis via JAK/STAT3 signaling | COSMIC, ClinVar |
| Infertility | LIF deficiency impairs embryo implantation | OMIM, NCBI |
| Inflammatory disorders | LIF modulates immune responses and cytokine production | UniProt, NCBI |
| Neurological diseases | LIF supports neuronal survival and regeneration | NCBI, UniProt |

Protein Summary
Leukemia inhibitory factor (LIF) is a secreted glycoprotein of 202 amino acids (UniProt P15018). It binds to the LIF receptor (LIFR) and gp130, activating JAK/STAT3 and MAPK pathways. LIF maintains embryonic stem cell pluripotency, regulates hematopoietic differentiation, and modulates inflammatory responses. Overexpression is linked to several cancers.
