Description

LGALS8 encodes galectin-8, a member of the galectin family of beta-galactoside-binding proteins. Galectin-8 contains two carbohydrate recognition domains (CRDs) connected by a linker peptide. It functions in cell adhesion, migration, immune modulation, autophagy, and endocytosis. LGALS8 is widely expressed and its dysregulation is implicated in cancer, autoimmune diseases, and infectious diseases.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Cancer (various types) Altered expression of galectin-8 modulates tumor cell adhesion, migration, and immune evasion PMID: 25687249; PMID: 29127120
Rheumatoid arthritis Galectin-8 promotes inflammatory cytokine production and osteoclastogenesis PMID: 28336555
Systemic lupus erythematosus Galectin-8 levels correlate with disease activity and autoantibody production PMID: 29562367
Infectious diseases (e.g., Salmonella, HIV) Galectin-8 restricts bacterial and viral infection by targeting pathogen-containing vacuoles to autophagy PMID: 22980980; PMID: 26068852

Protein Summary

Galectin-8 is a 359-amino acid protein with two CRDs (N-terminal and C-terminal) connected by a linker. It exists as a homodimer and binds beta-galactoside sugars on cell surface glycoproteins. Galectin-8 modulates cell adhesion by interacting with integrins, regulates autophagy by recognizing damaged endosomes, and influences immune responses by activating dendritic cells and T cells. Its expression is altered in various cancers and inflammatory diseases.
