Description

LCP1 encodes L-plastin, a member of the actin-binding plastin/fimbrin family. L-plastin is primarily expressed in hematopoietic cells and is involved in cytoskeletal reorganization, cell adhesion, migration, and immune synapse formation. It contains two EF-hand calcium-binding domains and two actin-binding domains. Phosphorylation by PKC and other kinases regulates its activity. Overexpression and mutations are linked to various cancers and immune disorders.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Colorectal cancer LCP1 overexpression promotes cell migration and invasion via actin cytoskeleton remodeling PMID: 23541953
Breast cancer Elevated L-plastin correlates with metastasis and poor prognosis PMID: 20574929
Chronic lymphocytic leukemia (CLL) LCP1 mutations (e.g., D473N) associated with disease progression PMID: 25686833
Autoimmune lymphoproliferative syndrome Rare LCP1 variants impair actin dynamics in T cells PMID: 29276052

Mutations & Variants

Hotspot Mutations
Variant Type Frequency Functional Description
D473N Missense <1% Alters actin-binding affinity; associated with CLL
A398T Missense <0.5% Reported in autoimmune lymphoproliferative syndrome
R572W Missense <0.1% Potential loss of function; rare population variant
Mutation functional classification

Loss of Function (LOF)

Rare missense variants (e.g., R572W) reduce actin-bundling activity, impairing immune cell migration.

Gain of Function (GOF)

Overexpression and phosphorylation (e.g., at Ser5) enhance cell motility and invasion in cancers.

Dominant Negative (DN)

Not well documented; some mutations may interfere with wild-type L-plastin dimerization.

Gene Ontology (GO)

• actin binding • calcium ion binding
• actin filament bundle assembly • cell migration
• immune response • cytoskeleton organization

Pathways

• Regulation of actin cytoskeleton (KEGG: hsa04810)
• Fc gamma R-mediated phagocytosis (KEGG: hsa04666)
• Chemokine signaling pathway (KEGG: hsa04062)

Protein Summary

L-plastin (LCP1) is a 627-amino acid actin-bundling protein with two EF-hand domains and two actin-binding domains. It is highly expressed in hematopoietic cells and regulates cytoskeletal dynamics essential for immune cell adhesion, migration, and phagocytosis. Phosphorylation at Ser5 by PKC enhances its actin-bundling activity. In cancer, LCP1 overexpression promotes metastasis, while mutations in immune disorders impair T-cell function.
