Description
LCN2 encodes lipocalin 2, also known as neutrophil gelatinase-associated lipocalin (NGAL). This secreted protein binds to bacterial siderophores, thereby sequestering iron and exerting bacteriostatic effects. It is involved in innate immunity, iron homeostasis, inflammation, and cellular stress responses. LCN2 is widely used as a biomarker for acute kidney injury and is implicated in various cancers, metabolic disorders, and inflammatory diseases.

Lipocalin 2 (NGAL) is a 25 kDa secreted glycoprotein belonging to the lipocalin family. It adopts a typical β-barrel structure that forms a ligand-binding pocket. Its primary function is to bind bacterial siderophores (e.g., enterobactin), thereby preventing bacterial iron acquisition. In mammals, LCN2 also binds to endogenous siderophores (catechol) and modulates cellular iron delivery. It forms a complex with matrix metalloproteinase-9 (MMP-9), protecting it from degradation and enhancing its activity. LCN2 is expressed in neutrophils, renal tubular cells, hepatocytes, and epithelial cells, and its levels rise dramatically in response to infection, inflammation, and tissue injury.
