Description
LARP1 encodes a member of the La-related protein family, characterized by a La motif and an RNA recognition motif. The protein functions as an RNA-binding protein that regulates the translation and stability of mRNAs, particularly those encoding ribosomal proteins and translation factors. It is a downstream effector of the mTORC1 signaling pathway and plays a critical role in ribosome biogenesis and cell growth. LARP1 is overexpressed in various cancers and is associated with poor prognosis.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Cancer (multiple types) | Overexpression of LARP1 promotes translation of mRNAs involved in cell proliferation and survival, contributing to oncogenesis. | NCBI Gene, COSMIC |
| Hepatocellular carcinoma | LARP1 upregulation enhances ribosome biogenesis and tumor growth via mTORC1 signaling. | PubMed, COSMIC |
| Colorectal cancer | LARP1 overexpression correlates with advanced stage and poor survival, likely through enhanced translation of oncogenic mRNAs. | PubMed, COSMIC |

Mutation functional classification
Loss of Function (LOF)
Nonsense and frameshift mutations that truncate the protein, likely impairing RNA binding and translational regulation.
Gain of Function (GOF)
Not well characterized; overexpression in cancer may act as a gain-of-function mechanism.
Dominant Negative (DN)
No dominant-negative mutations reported.

Protein Summary
LARP1 is a 1096-amino acid RNA-binding protein containing an N-terminal La domain and a C-terminal RNA recognition motif. It binds to the 5' terminal oligopyrimidine (TOP) motif of mRNAs encoding ribosomal proteins and translation factors, promoting their translation in response to growth signals. LARP1 is phosphorylated by mTORC1 and S6K, linking nutrient signaling to ribosome production. Its overexpression in tumors suggests a role in cancer cell growth and survival.
